Brief Method Summary
1 mL of sample is added to plate and swirled. Plate is allowed to incubate at room temperature for 20 minutes, then incubated at 35C for 48 hours. Following 48 hours incubation, number of colonies are counted.
Scope and Application
The EasyDisc PCA method is used for the quantification of culturable, waterborne bacteria. Culturable is defined as any aerobic bacteria able to form colonies on solid media.
Applicable Concentration Range
< 300
Interferences
Chlorinated samples should be treated with sodium thiosulfate prior to testing, preferably upon sample collection.
Quality Control Requirements
. The following quality control procedures are recommended for each lot of EasyDisc PCA: a. Positive Control option 1: IDEXX-QC HPC/TVC2 : Enterococcus faecalis b. Positive Control option 2: Fill a sterile vessel with 100 mL of sterile, non-buffered, oxidant-free water and inoculate with a sterile loop of any of the following strains: Enterococcus faecalis WDCM# 00087; ATCC# 29212 Escherichia coli WDCM# 00090; ATCC#11775 Negative Control/Blank test plates should not contain any colonies
Sample Handling
Maintain aseptic protocol
